If you connect somewhere, the relevant number belongs to the leg that lands in the country, not the first departure of the day. Your boarding pass or the airline confirmation email carries the number in the correct format, carrier letters included.
The e-Ticket is not only for flights. It also applies to cruise and other maritime arrivals. The busiest airports are Punta Cana (PUJ), Las Americas in Santo Domingo (SDQ), Cibao in Santiago (STI), Puerto Plata (POP), La Romana (LRM) and Samana (AZS).
